Evaluation of restricted access media for the purification of cell culture‐derived Orf viruses

Recently, multimodal chromatography using restricted access media (RAM) for the purification of nanoparticles, such as viruses has regained increasing attention. These resins combine size exclusion on particle shell and adsorptive interaction within core. Accordingly, smaller process-related impurities, example, DNA proteins, can be retained, while larger product pass unhindered. We evaluated a range currently available RAM, differing in shells’ pore cut-off core chemistry, cell culture-derived clarified model virus, namely Orf virus (ORFV). examined impurity depletion recovery relevant criteria evaluation column performance, well scale-up robustness regeneration potential evaluating multiple use application. The results indicate that some columns, Capto Core, enable both high protein removal, others, Monomix Core 60 (MC60), are more suitable depletion. Furthermore, is facilitated by columns with pores (5000 vs. 700 kDa) weaker binding interactions (anion exchange multimodal). According to these findings, choice RAM should selected according respective feed sample composition planned number application cycles.
